In this study, we sought to provide a comprehensive overview of CRESTOR’s cardiovascular benefit and impact, and to analyze the potential risk-benefit profile of this drug in the long term. We identified all patients with stable or uncontrolled LDL-c levels of <100 mg/dL who had stable LDL-c levels over 1.0 mmol/L and who were eligible for CRESTOR. Methods

This was a retrospective review of controlled clinical trials that were retrieved from MEDLINE, Embase, and the Cochrane Central Register of Controlled Trials (CENTRAL) using the terms CRESTOR and statin, and in combination with other drugs. A total of 1036 patients were included in the study. The primary outcome was the rate of a statin-associated cardiovascular event (ASC). The secondary outcome was the incidence of the event over a 5-year period, for each of the 5-year period from the last observation carried forward (LOCF), the primary endpoint (ASC) and the secondary endpoints (ASC and CHD).

Results

Study Characteristics

In total, 822 patients were identified as having stable or controlled LDL-c (≥100 mg/dL) who were eligible for CRESTOR. The median time to event was 4.2 years, and the median duration of LDL-c control was 3.3 years. We identified 632 patients who received CRESTOR for the first 6 months of treatment, and 533 patients who received a placebo arm at the first time point of the first event. We identified 615 patients who had stable LDL-c levels over 1.0 mmol/L and were eligible for CRESTOR.
